Anti-Treslin, clone 5L19 ZooMAb, Cat. No. ZRB1063, is a recombinant Rabbit monoclonal antibody that specifically targets Treslin and is tested for use in Affinity Binding Assay, Immunocytochemistry, and Western Blotting.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Treslin in NIH 3T3 cells. Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Target description
Treslin (UniProt: Q7Z2Z1; also known as TopBP1-interacting checkpoint and replication regulator, TopBP1-interacting, replication-stimulating protein) is encoded by the TICRR (also known as C15orf42) gene (Gene ID: 90381) in human. Treslin is a well conserved nuclear protein that associates with replicating chromatin. It serves as a regulator of DNA replication by promoting integration of Cdc45 into the replicative helicase. It is a key positive-regulatory target of cell-cycle control mechanisms and activation of Treslin by cyclin-dependent kinase (CDK) is shown to be essential for the initiation of replication. It possesses an unusually large number of potential CDK phosphorylation sites. Checkpoint-regulatory kinase Chk1 is shown to associate with a C-terminal domain of Treslin (TRCT) and mutations in the TRCT domain are known to abolish binding of Chk1 to Treslin and thereby eliminate its phosphorylation by Chk1. Disruption of the Treslin-Chk1 interaction is shown to increase initiation of chromosomal DNA replication. Two isoforms of Treslin have been described that are produced by alternative splicing. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.
